In this case study, L-α-Phosphatidylcholine extracted from soybeans (soyPC) was used to modify a methacrylate-based monolithic column in a capillary format via 1-ethyl-3-(3-dimethylaminopropyl) carbodiimide coupling, creating a stationary phase suitable for simulating cell surface membranes. The method for functionalizing the poly(GMA-co-EDMA) monolith with SoyPC is as follows:
The capillaries containing the monolithic substrate were initially flushed with water, followed by 2 M aqueous HMD solution at 60℃ for 2 hours at a flow rate of 60 μL/h. Next, the column was filled with 1 M aqueous GA solution, sealed with septum caps, and left at room temperature for 12 hours. Afterward, the column was flushed with water and methanol (MeOH). The modification step with 2 M HMD solution was repeated, and subsequently, the column was filled with a mixture containing 100 mM EDC and 150 mM soyPC dissolved in 0.1 M imidazole in MeOH. The column was sealed with septum caps and allowed to react for 30 minutes at room temperature. Following this, the column was filled with an aqueous solution of 0.2 M SCBH and left to react for 2 hours at 4℃. Finally, the column was washed with MeOH and isopropanol (IPA) to eliminate any soyPC adsorbed to the monolith through Van der Waals interactions.

In this case, L-α-glycerylphosphorylcholine (L-α-GPC) was prepared by catalytic hydrolysis of soybean PC (L-α-phosphatidylcholine) using immobilized Lecitase Ultra (LU). The preparation method is as follows:
The reaction is conducted in a flat-bottom glass container equipped with a water jacket to control the temperature. The container is preheated to the desired temperature using a water circulator. Then, 4 g of soybean PC is mixed with 16 mL of n-hexane, the required volume of water, and an immobilized enzyme. During the reaction, the mixture is stirred with a magnetic stirrer at a frequency of 10 Hz.
L-α-GPC is purified through phase separation in a biphasic medium of n-hexane and water. The reaction product is filtered under vacuum through Whatman No. 41 filter paper to remove the immobilized enzyme. The reaction vessel is washed with water and filtered as described above. The combined filtrate and washings are mixed with water and n-hexane in a separatory funnel to dissolve L-α-GPC and free fatty acids, respectively. After phase separation, the bottom aqueous layer is collected, dried in a rotary vacuum evaporator at 60°C, and further dried under nitrogen purging to obtain L-α-GPC.
