A Case Study on the Direct Antimycobacterial Effect of PAF C-16

This study investigated the direct impact of exogenous PAF C-16 on the growth of Mycobacterium tuberculosis, using M. bovis BCG and M. smegmatis as models of tuberculous mycobacteria. The experimental steps are as follows:
M. smegmatis:
Liquid cultures are grown in Luria-Bertani (LB) broth with 50 μg/ml carbenicillin, 0.15% (v/v) glycerol, and 0.1% (v/v) Tween-80. Incubated in a shaking incubator at 37℃ until the optical density (O.D) at 600 nm reached 0.8-0.9.
Colony-forming units (CFUs) per ml are determined by plating various dilutions of the bacterial stock on LB agar plates in triplicates and counting CFUs after 72 hours of incubation at 37℃.
M. bovis BCG:
Liquid cultures are grown in Middlebrook 7H9 broth supplemented with 10% (v/v) albumin dextrose catalase (ADC) and 0.2% (v/v) Tween-80. ADC is prepared by dissolving 5 g of bovine albumin fraction V, 2 g dextrose, 0.85 g sodium chloride, and 4 mg catalase from bovine liver in 100 ml of sterile water, then filtering the solution through a 0.22 μm filter.
Cultures are incubated in a shaking incubator at 37℃ for 2-3 weeks until the O.D at 600 nm reached 0.8-0.9. Plated various dilutions of the bacterial stock on 7H10 plates in triplicates and counting CFUs after 2-3 weeks of incubation at 37℃.
The study demonstrated that PAF C-16 inhibited the growth of M. bovis BCG and M. smegmatis in a dose- and time-dependent manner.
